Amalie Hassenplug was the youngest daughter of the civil servant Johannes Hassenpflug (1755-1834). Her mother Marie Magdalena Dresden came from a wealthy Huguenot family from Hanau. Her brother Ludwig Hassenpflug became a brother-in-law of the Brothers Grimm by marrying their sister Charlotte Grimm in 1822. Amalie contributed to several stories written by the Brothers Grimm.

Through the Brothers Grimm she moved in the Bökendorf Romantic circle around the Haxthausen family and there she first met Annette von Droste-Hülshoff in 1817. In 1837 they became close friends and Amalie had a strong influence on Annette's literary work in 1838 and 1839. After 1840 their friendship was less intensive, but it lasted until Annette's death in 1848. She also met Gretchen Verflassen, the head of an orphanage and a friend of Clemens Brentano. Gretchen looked after her mother until her death in 1840. But she was ill herself and died in 1845. Amalie wrote a sympathetic biography about her that was published in 1870 under the pseudonym AH.

In 1842 she moved to Berlin where she lived with her brother Ludwig. From 1845 to 1850 she lived in Hannover, where her friend Anna von Arnswaldt (born Haxthausen) lived as well. In 1848 Karl Christian Andrae painter her portrait. In 1866 she moved to Meersburg Castle, where she lived with Hildegard and Hildegund, the twin daughters of Annette's sister Jenny. She died in Meersburg and was buried next to Annette.
